Tritium labeling of the PXR ligand GW5801 - an unexpected tritium exchange reaction

Abstract

H-T exchange on 4,3,5-HO(Me3C)2C6H2CHO in presence of Crabtree's catalyst gave exclusively 4,3,5-HO(Me3C)2C6H2CTO, which was treated with CH2[P(O)(OEt)2]2 to give 4,3,5-HO(Me3C)2C6H2CT:CH[P(O)(OEt)2]2. The absence of ring tritiation in 4,3,5-HO(Me3C)2C6H2CHO is attributed to steric hindrance by the tert.-Bu groups. [on SciFinder(R)]
