Abstract
Antibody-drug conjugates (ADCs) are complex immunotherapies requiring consistent quality assessment from early discovery. Despite the development of robust large-scalemethods, microgram to milligram scale reaction scalability is underexplored.
Here, we define this using DAR, purity, hydrophobicity, and aggregation metrics via twomodel ADCs: a trastuzumab-dansyl dye mimic (T-dansyl) and a trastuzumab-JQ1
conjugate (T-JQ1) at 0.1–1.7 mg, across two reduction states (low and high DAR targeting).
